The SRY (Sex-determining Region Y) gene is crucial for determining male sex and is located on the Y chromosome. It directs the formation of testes by encoding the testis-determining factor (TDF), which activates genes essential for testis development. Without the SRY gene, the default development is female.

Genetic testing for the SRY gene is important in diagnosing disorders of sex development (DSDs). For instance, Swyer syndrome occurs when individuals with XY chromosomes develop as females due to mutations in the SRY gene. Conversely, XX male syndrome can happen when the SRY gene is translocated to an X chromosome, causing individuals with two X chromosomes to develop male characteristics.

This test is also valuable in prenatal care, helping determine the sex of the fetus and identify potential DSDs early, especially if there is a family history of these conditions. For infants with ambiguous genitalia, testing provides clarity on their sex and guides treatment decisions.

Overall, SRY gene testing plays a critical role in understanding and managing sex development, aiding in accurate diagnosis, treatment planning, and genetic counseling. It ensures informed decisions and personalized care, enhancing our approach to genetic conditions and prenatal health.
